Tricine-SDS-PAGE Protocol

Tricine-SDS Page Protocol

The following reagents are needed in advance:

Gel monomer (30% acrylamide, 29:1). 29.0 g acrylamide and 1.0 g bisacrylamide are dissolved in a total volume of 100 mL MQ H2O. Store at 4 °C, protect from light.

Gel Buffer. Dissolve 181.7 g Tris base and 1.5 g SDS in 400 mL MQ H2O. Adjust the pH to 8.45 using concentrated HCl. Bring to a final volume of 500 mL with MQ H2O. Store at 4 °C.

Anode Buffer (Lower Buffer). Dissolve 12.11 g Tris base in 400 mL MQ H2O. Adjust to pH 8.9 with HCl. Bring to 500 mL with MQ H2O. Store at 4 °C

Cathode Buffer (Upper Buffer). Dissolve 6.055 g Tris base and 8.96 g Tricine in a total volume of 500 mL MQ H2O. Add 0.5 g SDS. Store at 4 °C.

NOVEX 2X Tricine-SDS sample buffer. For 10mL, mix the following:

3.0 mL 3.0 M Tris-HCl, pH 8.45
2.4 mL glycerol
0.8 g SDS
1.5 mL 0.1% Coomassie Blue G
0.5 mL 0.1% Phenol Red
Distilled water to 10 mL
Other Reagents Needed: TEMED (pure liquid), 10 % (w/v) ammonium persulfate (made freshly), 0.1% (w/v) SDS, and glycerol.

Gel Preparation

Prepare & Pour the Separating Gel: Note - enough for four Novex 1.0 mm mini-gel cassettes. This recipe is for a 12.0 % running gel. In a disposable 50 mL conical tube, mix the following:

12.0 mL of gel monomer.
10 mL of gel buffer
3.1 mL glycerol
4.8 mL MQ H2O
0.1 mL 10% ammonium persulfate.
Iniatiate polymerization by adding 40 uL of TEMED. Transfer to the gel cassettes; bring to approximately 1.5 cm from the top of gel blank (which is reserved for the stacking gel) and gently overlay with a small volume (ca. 0.5 mL) of 0.1% SDS. Polymerize for 30 min. Remove layer of SDS after the gel has polymerized.

Prepare & Pour the Stacking Gel: Note - enough for four Novex 1.0 mm mini-gel cassettes. This recipe is for a 4.0 % stacking gel. In a disposable 50 mL conical tube, mix the following:

1.25 mL of gel monomer
2.5 mL of gel buffer
6.25 mL H2O
0.05 mL 10% ammonium persulfate
Iniatiate polymerization by adding 20 uL of TEMED. Transfer to the gel cassettes and insert comb. Clamp along the top with binder clips to insure well formed wells. Polymerize for 30 min.

Running, Developing and Drying the Gel.

Sample Preparation Samples are prepared by mixing a equivalent volume of sample with an equivalent volume of NOVEX 2X Tricine-SDS sample buffer. The samples are then heated for 5 min in a boiling water bath, centrifuged for one minute,

Gel Setup and Run Parameters. Prior to loading, fill the upper buffer chamber with cathode buffer and the lower chamber with 250 mL of anode buffer. Electrophorese at constant current - 15 mA until the samples have migrated through the stack; 30 mA for the remainder of the run (until the tracking dye has migrated out from the lower slot).

Staining. Staining is accomplished by soaking the gel in a solution of 45% methanol, 45% deionized water, 10% acetic acid, and 0.25% (w/v) Coomassie Blue R-250 on a table top shaker for 30 minutes. The gel is then destained by soaking in a solution of 45% methanol, 45% deionized water, and 10% acetic acid on a table top shaker. Periodically replace with fresh solution until desired destaining is achieved.

Gel Drying. After destaining, the gel is prepared for drying by soaking it for five minutes in gel drying solution (5% glycerol/30% methanol). Next, soak a single piece of cellophane in the same gel dry solution for 30 seconds. Lay down the cellophane first, and the gel on top of this. Next soak the second piece of cellophane in gel dry solution for 30 seconds, and then overlay this on the gel to make a sandwich. Squeeze out any air bubbles and then clamp on upper frame. Dry at room temperature.

      Stem Cell Research Rules in UK

      Britain to Decide on Stem Cell Research

      LONDON (AP) — Britain is considering whether to permit research using animal eggs to create human stem cells — a process that tests the ethical boundaries of cutting-edge scientific research.

      The Human Fertilization and Embryology Authority will rule Wednesday on whether to accept such research, which involves placing human DNA into cow or rabbit eggs that have had their genetic material removed. If the authority allows such research, each project will be decided on a case-by-case basis.

          RIO DE JANEIRO, Sept. 5 (Xinhua) -- The U.S. Monsanto Company announced on Wednesday it will invest 28 million U.S. dollars to develop a new type of transgenic soybean in Brazil resistant to the herbicide glyphosate.

          The U.S. company said its new transgenic soybean will be resistant to both glyphosate and the soybean worm (Anticarsia gemmatallis) and it will be the first time that such technology is developed in markets outside the United States.

          Monsanto's investment will be channeled into three research stations in the southern states of Parana and Rio Grande do Sul as well as in the northern state of Tocantins.

          Alfonso Alba, the company's president in Brazil, said that "the headway achieved in the regulatory process and the respect toward intellectual property rights in the country have led the company to increase its investments in Brazil."

      FORT LAUDERDALE, Fla., Sept. 5 /PRNewswire-FirstCall/ -- Imaging Diagnostic Systems, Inc., a pioneer in laser optical breast cancer imaging systems, has installed a CT Laser Mammography (CTLM(R)) system at the Tianjin Medical University's Cancer Institute and Hospital, the largest breast disease center in China.

      Research at the Cancer Institute will be led by Professor Bao Runxian, Director of Radiology. Data will be collected under three research protocols designed to improve current methods of addressing breast cancer imaging and treatment follow-up. Professor E. Milne, MD, IMDS Director of Clinical Research will train the University doctors on CTLM adjunctive image interpretation and protocol methodologies.

      The Tianjin system is the second research system installed in China. The first, at Beijing's Friendship Hospital, enabled CTLM clinical procedures to become listed on the Regional schedule for patient payments. IMDS plans to install several more research CTLM systems in China to accelerate market adoption of the new laser breast imaging technique.

      Both the incidence and the number of deaths from breast cancer has tripled in Chinese women, making it the second most common cause of death from cancer for Chinese women. Early detection may provide women with better treatment options. Sources in China report that the incidence of breast cancer in has increased by at least 27% over the past decade and 40% of the Chinese women diagnosed with breast cancer die within five years because the cancer is already very advanced when it is diagnosed.

      The Cancer Institute and Hospital is planning the 4th Congress of the World Society for Breast Health, in Tianjin, October 18th through October 21st. The Congress is sponsored by the World Society for Breast Health, the China Oncology Society and the Chinese Medical Association. IDSI Beijing will present CTLM clinical findings at the meeting.

      Large scale gene transfer between organisms

      Large scale gene transfer between single-celled and multicellular organisms reported

      Wolbachia has on some occasions inserted almost its entire genome into species that it infects reported scientists at the J. Craig Venter Institute and the University of Rochester. This is the first example of large-scale horizontal gene transfer between single-celled and multicellular organisms. Although horizontal gene transmission is common among single-celled organisms, it is rare among multicellular organisms, and large scale transfer like that of an entire genome had previously not been suspected.

      The scientists found that in addition to Wolbachia engaging in almost complete genome transfer into Drosophila ananassae, it also had made significant transfer in 3 other insects species and 4 species of nematodes. The researchers found candidate species by scanning genetic databases for sequences found in Wolbachia. The scientists also found that these added sections were conserved by reproduction; that is the added sections stayed in the genomes after multiple generations. Moreover, there is evidence that suggests that the segments of Wolbachia's genome increased the reproductive fitness of the insect species.

      The transfers likely occurred during attempts at DNA repair in which the repair mechanisms incorporated Wolbachia DNA (available since the cells were infected with Wolbachia) into the genomes. These results could have major implications for understanding of evolution. The research also has implications for various forms of sequencing research, since when sequencing species, bacteria sequences are frequently ignored as they are generally assumed to be contaminants rather than good data.

      Wolbachia, a genus of bacteria that normally infects anthropods, especially insects, is already known for its odd behavior that can affect species in strange ways. For example, Wolbachia has been shown to be correlated with fast evolution among species it infects and is suspected for being responsible for a variety of speciation events as a side affect of Wolbachia creating reproductive barriers. Wolbachia by some estimates infects more than half of all anthropods and is already thought to play a major role in the evolution and speciation of many invertebrates.

      Since Wolbachia can generally only reproduce through females, it has adopted a number of strategies that treat males and females of species differently that can result in reproductive barriers. These strategies include killing of males, forced parthenogenesis, and preventing infected males from reproducing with uninfected females.
